OBJECTIVE: To identify DNA methylation patterns of heavy smokers in oral rinse samples. METHODS: Genome-wide DNA methylation data was imported from Gene Expression Omnibus GSE70977 using the GEOquery package. Two independent sets were analyzed: (a) 71 epigenomes of cancer-free subjects (heavy smokers n = 37 vs. non-smokers n = 31); for concordance assessment (b) 139 oral-cancer patients' epigenomes (heavy smokers n = 92 vs. non-smokers n = 47). Differential DNA methylation for CpG positions and at the regional level was determined using Limma and DMRcate Bioconductor packages. The linear model included sex, age, and alcohol consumption. The statistical threshold was set to p < 0.05. Functional gene prioritization analysis was performed for gene-targeted analysis. RESULTS: In individuals without cancer and heavy smokers, the FAM184B gene was found with two CpG positions differentially hypermethylated (p = 0.012 after FDR adjustment), in a region of 48 bp with an absolute methylation difference >10% between groups (p = 1.76 × 10-8). In the analysis corresponding to oral-cancer patients, we found AHRR differentially hypomethylated cancer patients, but also in subjects without oral cancer in the targeted analyses. Remarkably, ADAMTS2 was found differentially hypermethylated in heavy smokers without a diagnosis of cancer in two consecutive probes cg05575921 (p = 3.13 × 10-7) and cg10208897 (p = 1.36 × 10-5). CONCLUSIONS: Differentially methylated AHRR, ADAMTS2, and FAM184B genes are biomarker candidates in oral rinse samples.

Introducción: evaluar la satisfacción con los tratamientos requiere cuestionarios específicos y validados que garanticen una prestación del servicio adecuada y que contribuya al bienestar de las personas. El objetivo fue evaluar la validez y la confiabilidad de la versión colombiana de un cuestionario sobre satis-facción en una población de usuarios de prótesis total. Materiales y métodos: se realizó un estudio de evaluación de tecnología diagnóstica con una población de 226 pacientes, a partir de las características sociodemográficas y los diez ítems del instrumento. Se analizó la consistencia interna, la reproducibilidad prueba-reprueba y los límites de acuerdo de Bland y Altman. Así mismo, se analizaron los componentes y se aplicó el modelo de Rasch. Resultados: participaron 158 (69.91 %) mujeres y la mediana de edad fue de 69 años. Se obtuvieron un alfa de Cronbach de 0.90 y un coeficiente de correlación intraclase de 0.91. Se identificaron dos factores, los diez ítems se ajustaron al modelo de Rasch y no se observó un funcionamiento diferencial del ítem. Conclusiones: la versión colombiana del cuestionario presentó excelente confiabilidad y las medidas obtenidas explicaron el 58.60 % de la varianza.


Introduction: Evaluating treatment satisfaction requires a specific and validated questionnaire that can obtain information about adequate service provision and contribute to people's well-being. The objective of this study was to evaluate the validity and reliability of the Colombian version of a questionnaire on satisfaction among complete denture wearers. Materials and methods: This survey evaluated diagnostic technology used among 226 patients. Information on sociodemographic characteristics was obtained and ten items of the instrument were evaluated. Internal consistency, test-retest reproducibility, and Bland-Altman limits of agreement were analyzed. Component analysis was performed and the Rasch model was used. Results: Median age of the participants was 69 (ric: 63-76) years, and 158 (69.91%) were women. Cronbach's alpha and intraclass correlation coefficients were 0.91 and 0.80, respectively. Two factors were identified, all ten items conformed to the Rasch model and no differential item functioning was observed. Conclusions: The Colombian version of the questionnaire showed excellent reliability, and the proportion of variance explained by the measures was deemed satisfactory (58.60%).

A consistência interna, a reprodutibilidade teste-reteste e os limites de concordância foram analisados de acordo com Bland e Altman. Foi reali-zada uma análise de componentes e aplicado o modelo Rasch. Resultados: participaram 158 (69,91%) mulheres, a mediana de idade foi de 69 anos. Obteve-se um alfa de Cronbach de 0,90 e um coeficiente de correlação intraclasse de 0,91. Dois fatores foram identificados, os dez itens se ajustaram ao modelo Rasch e nenhum funcionamento diferencial dos itens foi observado. Conclusões: A versão colombiana do questionário apresentou excelente confiabilidade e as medidas obtidas explicaram 58,60% da variância.

Background: Periodontal disease is considered as a diabetes complication and has been suggested that periodontal treatment plus antibiotics should reduce glycated hemoglobin A, by reducing local production of pro inflammatory substances. Objective: To evaluate diabetic patients with periodontal disease under periodontal treatment plus topical antibiotics and reduction of HbA1c, compared to diabetic patients under periodontal treatment without antibiotics. Materials and Methods: Using PUBMED, SCOPUS, WEB OF SCIENCE, EMBASE and Google Scholar data bases, were screened documents from 2008 to 2018. The documents included were the clinical studies, which included non-surgical periodontal treatment plus topical antibiotics, whose outcomes included the HbA1c report. Two independent researchers evaluate title; abstract and bias risk with Downs Black scale and Cochrane tool. Documents with a score higher than 15 on average by the two evaluators were included. Results: Five articles, which find inclusion criteria, were identified. Two documents failed to demonstrate statistically significant effect when compared to non-surgical periodontal therapy alone. Conclusion: In general a modest reduction of HbA1c was identified when using antibiotic therapy.


Antecedentes: La enfermedad periodontal es considerada una complicación del paciente diabético y se ha sugerido que su tratamiento con o sin antibióticos podría aportar a la reducción de la hemoglobina glucosilada, al reducir la producción local de sustancias pro-inflamatorias. Objetivo: Evaluar pacientes diabéticos con enfermedad periodontal que recibieron terapia periodontal no quirúrgica (TPNQ) más antibióticos tópicos y la reducción de la hemoglobina glucosilada, comparados con pacientes que recibieron TPNQ sin antibióticos tópicos. Materiales y métodos: Se utilizaron bases de datos MEDLINE vía PubMed, WEB OF SCIENCE, SCOPUS, EMBASE y Google Scholar utilizando vocabulario controlado y limitando la búsqueda a los años 2008-2018. Los documentos incluidos fueron los estudios clínicos, que incluían tratamiento periodontal no quirúrgico mas antibióticos tópicos, cuyos desenlaces incluyeran el reporte de HbA1c. Estos fueron analizados por dos evaluadores independientes, para identificación de riesgo de sesgo con la escala Downs Black y herramienta Cochrane. Se incluyeron los documentos con puntuación mayor a 15 en promedio de los dos evaluadores. Resultados: Se identificaron 5 documentos que cumplen con los criterios de inclusión. Dos documentos no mostraron diferencias estadísticamente significativas para el cambio de la HbA1c, en comparación con la terapia periodontal sola. Conclusión: En general se evidencia una discreta reducción de HbA1c con la terapia periodontal usando antibióticos tópicos.

Aim: To determine the relationship between the chemical composition of saliva, periodontal disease and dental calculus. Methods: An observational analytical cross-sectional study was conducted with patients over 55 years of age. Ethical principles of autonomy and risk protection were applied according to the international standards. Sociodemographic and diagnosis variables (presence of dental calculus and periodontal status) were considered to measure salivary concentrations of glucose (by the glucose oxidase/peroxidase method, amylase (by the colorimetric test), urea (by the amount of indophenol), total protein (by the Bradford method) and albumin (by the nephelometric method). Patients chewed a sterile rubber band and 3 mL of stimulated saliva were collected. The samples were stored at -5 °C, centrifuged at 2,800 rpm for 10 min, and the supernatant was removed and stored at -20 °C. Data were presented as frequencies and proportions for qualitative variables and measures of central tendency and dispersion for quantitative variables. Data were analyzed by either analysis of variance or Kruskal Wallis test . A p value <0.05 was considered statistically significant. Results: Significant relationships were observed between the concentration of salivary urea and periodontal status (p = 0.03) and the presence of dental calculus and urea (p = 0.04) was demonstrated. Conclusions: A relationship between the salivary urea concentration and the presence of periodontal disease and dental calculus is suggested.
